Azithromycin dihydrate
Indications/Uses
Resp tract infections, otitis media, skin & soft tissue infections, uncomplicated genital chlamydial infections & non-gonococcal urethritis, mild or moderate typhoid due to multiple antibacterial resistant organisms. Prophylaxis of endocarditis in childn.
Dosage/Direction for Use
Adult 500 mg as single dose daily for 3 days. Alternatively, initially 500 mg followed by 250 mg daily for a further 4 days. Uncomplicated genital infection caused by Chlamydia trachomatis & chancroid 1 g as single dose. Uncomplicated gonorrhea 2 g as single dose. Granuloma inguinale Initially 1 g followed by 500 mg daily, or 1 g once wkly for at least 3 wk, until all lesions have completely healed. Prophylaxis of disseminated MAC infection 1.2 g once wkly. Treatment or secondary prophylaxis 500 mg once daily in combination w/ other antimycobacterials. Mild to moderate typhoid caused by multidrug-resistant strain 500 mg once daily for 7 days.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity to azithromycin, erythromycin, any macrolide or ketolide antibiotic.
Special Precautions
Discontinue use if allergic reaction occurs. Does not treat viral infections. Pseudomembranous colitis. Prolonged cardiac repolarization & QT interval, imparting a risk of developing cardiac arrhythmia & torsades de pointes. Not to be simultaneously taken w/ Al & Mg-containing antacids. Hepatic impairment.
Adverse Reactions
Nausea, vomiting, diarrhea, abdominal pain.
Drug Interactions
Reduced peak serum levels w/ Al & Mg-containing antacids. Increased serum conc of theophylline. Increased anticoagulant effects of warfarin. Elevated levels of digoxin. Acute ergot toxicity w/ ergotamine or dihydroergotamine. Decreased clearance & thus may increase pharmacologic effect of triazolam. Elevated serum levels of carbamazepine, cyclosporine, hexobarbital, & phenytoin.
